Compounds in mate tea induce death in colon cancer cells, in vitro study shows

ScienceDaily (Jan. 23, 2012) ? Could preventing colon cancer be as simple as developing a taste for yerba mate tea? In a recent University of Illinois study, scientists showed that human colon cancer cells die when they are exposed to the approximate number of bioactive compounds present in one cup of this brew, which has long been consumed in South America for its medicinal properties.

"The caffeine derivatives in mate tea not only induced death in human colon cancer cells, they also reduced important markers of inflammation," said Elvira de Mejia, a U of I associate professor of food chemistry and food toxicology.

That's important because inflammation can trigger the steps of cancer progression, she said.

In the in vitro study, de Mejia and former graduate student Sirima Puangpraphant isolated, purified, and then treated human colon cancer cells with caffeoylquinic acid (CQA) derivatives from mate tea. As the scientists increased the CQA concentration, cancer cells died as a result of apoptosis.

"Put simply, the cancer cell self-destructs because its DNA has been damaged," she said.

The ability to induce apoptosis, or cell death, is a promising tactic for therapeutic interventions in all types of cancer, she said.

de Mejia said they were able to identify the mechanism that led to cell death. Certain CQA derivatives dramatically decreased several markers of inflammation, including NF-kappa-B, which regulates many genes that affect the process through the production of important enzymes. Ultimately cancer cells died with the induction of two specific enzymes, caspase-3 and caspase-8, de Mejia said.

"If we can reduce the activity of NF-kappa-B, the important marker that links inflammation and cancer, we'll be better able to control the transformation of normal cells to cancer cells," she added.

The results of the study strongly suggest that the caffeine derivatives in mate tea have potential as anti-cancer agents and could also be helpful in other diseases associated with inflammation, she said.

But, because the colon and its microflora play a major role in the absorption and metabolism of caffeine-related compounds, the anti-inflammatory and anti-cancer effects of mate tea may be most useful in the colon.

"We believe there's ample evidence to support drinking mate tea for its bioactive benefits, especially if you have reason to be concerned about colon cancer. Mate tea bags are available in health food stores and are increasingly available in large supermarkets," she added.

The scientists have already completed and will soon publish the results of a study that compares the development of colon cancer in rats that drank mate tea as their only source of water with a control group that drank only water.

This in vitro study was published in Molecular Nutrition & Food Research, vol. 55, pp. 1509-1522, in 2011. Co-authors include Sirima Puangpraphant, now an assistant professor at Kasetsart University in Thailand; Greg Potts, an undergraduate student of the U of I; and Mark A. Berhow and Karl Vermillion of the USDA, ARS, National Center for Agricultural Utilization Research in Peoria, Illinois. The work was funded by the U of I Research Board and Puangpraphant's Royal Thai Government Scholarship.

'The Devil Inside' writers are making another movie -- a sequel?

'The Devil Inside' writers say they're making another found-footage horror movie. Will it be a 'Devil' sequel or something new?

Hot on the heels of a demonic monster opening weekend at the box office (to the tune of $34.5 million) for The Devil Inside, the film?s writer/director (William Brent Bell) and co-writer (Matthew Peterman)?are already preparing to shoot another, currently untitled, ?found footage? style horror film.

Skip to next paragraph Screen Rant

Screen Rant had a humble start back in 2003 as a place to rant about some of the dumber stuff related to the movie industry. Since then, the site has grown to cover more and more TV and movie news (and not just the dumb stuff) along with sometimes controversial movie reviews. The goal at Screen Rant is to cover stories and review movies from a middle ground/average person perspective.

Recent posts

It?s no surprise to hear that the pair is moving fast, especially considering that most box office experts are expecting a major drop-off for The Devil Inside in the coming week ? due to negative word of mouth and the unenthusiastic response of the critical community (read our review of The Devil Inside). However, regardless of the film?s reception, it managed to do big bucks on a $1 million budget ? making the pair an attractive prospect for Hollywood executives.

According to The Hollywood Reporter, CEO of Sierra/Affinity,?Nick Meyer?had no problem touting the pair?s accomplishments ? even if The Devil Inside legacy is still up in the air:

?From day one, we have been huge fans of this filmmaking team and the way they have crafted a unique movie-going experience with? The Devil Inside. These are filmmakers we know and trust and they bring an original voice and style to our next project that will once again introduce audiences to a new expectaiton of what one can define as a genre film.?

Bell and Peterman?will, as mentioned, write the film together with Bell once again pulling double-duties as director. The film is set for financing through Sierra/Affinity and the Incentive Filmed Entertainment Fund with Peterman as well as?Steven Schneider (Paranormal Activity, Insidious) and Morris Paulson (The Devil Inside) producing ? and Bell, Peterman, and Paulson?s company, Prototype, handling production. The film is?set to start work in Romania this April ? though no storylines or potential cast members have been revealed at this point.

Admittedly, this new project was just announced but it?s hard not to be somewhat suspicious given the lack of concrete details, and Schneider?s annualizing of the?Paranormal Activity series -?not to mention The Devil Inside cliffhanger and decent box office profit. Is it a possibility that, despite Meyer?s claims that the new film will set ?a new expectaiton of what one can define as a genre film,??that the new film?could be the no-brainer?Devil Inside sequel?

Of course, with all the uncertainty around whether or not audiences would be receptive to a Devil Inside 2, there?s always the possibility that the pair is actually preparing a different ?found footage? genre experience ? hopefully with a better ending this round. A Deadline report indicates that the new Bell/Peterman film will ?bring a similar filmmaking style to another classic horror mythology? ? though, with so few details to go on, it?s possible that the movie?s producers are merely trying to muddy the waters a bit and keep people from speculating that the new film is actually just another installment in Hollywood?s next big annualized cash grab.?We?ll likely know more next month ? as?Sierra/Affinity is expected to sell the project to potential foreign buyers at the European Film Market in Berlin.

Afghan government: cease-fire before Taliban talks (AP)

KABUL, Afghanistan ? Taliban insurgents must agree to a cease-fire before formal peace negotiations can begin in Qatar, a spokesman for Afghan President Hamid Karzai said Monday

Presidential spokesman Emal Faizi insisted that the government will never give up territory to the insurgents.

Faizi laid out the government's terms days after the Taliban's announcement it would open a political office in Doha, Qatar, a key precursor to peace talks and the insurgents' first public move toward a political settlement to the 10-year-long war.

"When the talks start, there should be a cease-fire and the violence against the Afghan people must stop," Faizi said Monday on Tolo television news.

He dismissed as "baseless" speculation in Afghan media that the government might hand over predominantly Pashtun southern provinces to the Taliban in exchange for an end to the fighting.

"The Islamic Republic of Afghanistan will never accept such suggestion from any side," he said

Faizi also said it is too soon to send a delegation to Qatar to discuss future talks.

Afghan High Peace Council member Mohammad Ismail Qasimyar said earlier Monday that the panel has asked the government to send a delegation to Doha "as soon as possible," but Faizi said the government has no immediate plans for such a trip.

The U.S. has been seeking to start negotiations as international troops begin to leave the country, according to plans to withdraw most foreign security forces by the end of 2014. Washington says any negotiations must be Afghan-led.

Debate over peace talks came as violence continued.

An American soldier was killed when man in an Afghan army uniform opened fire at a base in the south of the country, an Afghan military spokesman said Monday. Another American soldier was wounded in the Sunday attack. Earlier reports did not identify the nationality of the soldiers.

Spokesman Gen. Mohammad Zahir Azimi said the gunman was also killed in the shootout. "Right now, an investigation is going on to determine whether he really was a soldier or someone using an army uniform. And if he was a soldier, what caused the shooting," Azimi said.

Similar attacks have raised fears of increased Taliban infiltration of the Afghan police and army as NATO speeds up the training of the security forces. In some cases the attackers were Afghan soldiers who turned on NATO troops. Others involved insurgents dressed in Afghan uniforms.

The shooting brought to 11 the number of NATO soldiers killed this month.

NATO is encouraging the rapidly expanding Afghan security forces to take more responsibility ahead of the coalition's 2014 pullout target date. NATO's training mission hopes have about 350,000 Afghan troops and police trained and ready by then.

Also Monday, Taliban fighters ambushed a convoy carrying supplies for NATO in western Afghanistan, killing two private security guards and wounding three. Seven of the attacking insurgents were also killed, said deputy police chief Mohammad Ghaws Milyar.

Iran lacks avenues for condemning hits on scientists (Reuters)

UNITED NATIONS (Reuters) ? Iran may be outraged at the killing of another nuclear scientist in broad daylight, but it lacks viable avenues for international condemnation or prosecution of what could be an attempt to sabotage its nuclear program.

Tehran urged the U.N. Security Council and Secretary-General Ban Ki-moon on Wednesday to condemn the latest in a series of assassinations, which it said were "cruel, inhumane and criminal acts of terrorism" aimed at undermining a nuclear program that Western powers and Israel suspect is for weapons.

Iran insists its nuclear program is peaceful and has defied Security Council demands - backed up by four rounds of U.N. sanctions - that it stop enriching uranium.

Iran's U.N. Ambassador Mohammad Khazaee appealed to Ban and the 15-nation council "to condemn, in the strongest terms, these inhumane terrorist acts and to take effective steps towards elimination of terrorism in all its forms and manifestations."

"Any kind of political and economic pressures or terrorist attacks targeting the Iranian nuclear scientists, could not prevent our nation in exercising this right" to pursue its nuclear program, Khazaee said in a letter, obtained by Reuters.

The United Nations has not heeded previous Iranian calls for condemnations of similar assassinations. Even if the Security Council were to take up the issue, the United States could use its veto power to block even the mildest condemnation.

Ban's spokesman Martin Nesirky told reporters earlier on Wednesday that the United Nations was aware of the reports out of Tehran but had no immediate comment.

But Christof Heyns, the U.N. special rapporteur on extrajudicial executions, said in a statement to Reuters the Wednesday assassination seemed to reflect a "worrying trend of extrajudicial executions of nuclear scientists in Iran."

"The killings are unlawful and should be condemned," Heyns said. "The onus is on the Iranian authorities to investigate what has happened, to make the evidence known and to bring the perpetrators to book."

Tehran blamed the United States and Israel for the attack, although Khazaee left that out of his letter. The White House denied any U.S. role and Israel declined to comment.

"STICKY MAGNETIC BOMB"

"Based on the existing evidence collected by the relevant Iranian security authorities, similar to previous incidents, perpetrators used the same terrorist method in assassinating Iranian nuclear scientists," Khazaee said.

He said the assassins worked by attaching "a sticky magnetic bomb to the car carrying the scientists and detonating it."

In October 2011, Khazaee complained to the council and Ban Ki-moon about the hits on Iranian scientists in a letter responding to U.S. allegations that Iran had plotted to assassinate Saudi Arabia's ambassador to Washington.

In early November, Iran's Supreme Leader Ayatollah Ali Khamenei said he had 100 "undeniable documents" proving that Washington was behind the "terrorist acts" in Iran.

Saeed Jalili, the secretary of Iran's Supreme National Security Council, said Iran was handing over to the United Nations all the evidence of U.S. plots. Nothing came of the Iranian complaints.

Later in November, the overwhelming majority of the 193-nation U.N. General Assembly voted to condemn the alleged Iranian plot. Unlike the Saudis, Iran was unable to secure a condemnation of the assassinations of its scientists.

In the absence of war, targeted killings are illegal under international law. If Israel or the United States were found to be involved in the assassinations, it would violate the U.N. Charter, which bans the use of force against sovereign states.

Tehran could launch legal proceedings in Iran. The International Court of Justice in The Hague, established by the U.N. Charter, would be the most obvious international forum for Iran to bring a legal case. But the court will only hear cases where states involved jointly request its involvement.

One of the reasons the Saudi plot angered U.N. member states was that it was aimed at an envoy of a sovereign government. U.N. member states traditionally have no tolerance for threats against diplomats, whether friend or foe.

In 2010, Israel's Mossad intelligence agency was accused of sending a hit squad to assassinate a Hamas militant in a Dubai hotel. The suspected agents were caught on videotape following their target in the hotel.

Authorities in Britain and other European capitals launched their own investigations into the Dubai hit, but not because a member of Hamas had been killed. They were irritated by the fact that fraudulent European passports were used by the hit squad.
